KPV
KPV research includes cells, animal models and engineered delivery systems. The peptide form and the way it is delivered shape each result.
What is KPV?
KPV is shorthand for lysine, proline and valine, three amino acids at the end of alpha-MSH. Papers and products use the name for several chemical forms. The full sequence, end groups and salt distinguish those forms.1711,1712
Are KPV and KPV acetate the same thing?
The acetate form includes an acetate counterion. FDA evaluated it separately from the form it calls KPV free base. Their listed molecular weights are about 402.5 and 342.43 g/mol, respectively.1711,1713,1712
What does the NH2 at the end mean?
It marks an amide at the peptide’s C-terminus. H-Lys-Pro-Val-NH2 and H-Lys-Pro-Val-OH have different chemical structures and molecular weights. A paper using one form needs to be identified as such.1711,1714
Why do some papers use acetylated or D-amino-acid forms?
Researchers change peptide structures to study activity and breakdown. Adding an acetyl group or changing an amino acid’s configuration creates a different molecule. The formula alone may not distinguish forms with different stereochemistry.1715,1712
Is KdPT the same peptide?
The published ulcerative-colitis trial names its peptide K(D)PT. Its retained title and abstract do not expand that name into a full chemical sequence. The trial stays separate from studies naming KPV.1731
Why are the Chinese burn studies treated separately?
Their intervention was Ac-D-Lys-L-Pro-D-Val, an acetylated analogue with two D-amino acids. The abstracts report receptor changes after scald injury in rats. They leave dose, group size and the specific injection route unstated.1732,1733
What is (CKPV)2?
(CKPV)2 is a cysteine-linked dimer, written as (Ac-Cys-Lys-Pro-Val-NH2)2. It has extra cysteines, acetyl groups and amidated ends. Its studies are identified separately throughout this guide.1734
Why do so many KPV papers use nanoparticles or hydrogels?
Researchers use carriers to change delivery, protect the peptide or combine it with another treatment. KPV sometimes serves as a targeting ligand rather than the main active ingredient. Each result belongs to the complete formulation and its comparison groups.1735,1736,1737,1730
Evidence in people
Has KPV actually been studied in people?
No cited study administered exact KPV to people and measured treatment outcomes. FDA reported the same gap in its May 2026 review. Human-use claims at the July hearing still need appraisal of the underlying patient records and methods.1712,1716,1717
Why do studies mention human blood or skin?
Researchers exposed removed blood cells or skin to the peptide in the laboratory. Examples include blood from people with HIV and skin transport experiments. Treatment studies measure outcomes after giving a defined product to living participants.1718,1719
What about the claims of millions of doses?
At the July hearing, a speaker claimed retrospective data covering 3.66 million KPV doses. A person can receive many doses. The exhibit, event definitions and follow-up need review before a safety rate can be assessed.1716
Did the hearing establish that KPV works?
The hearing recorded clinical anecdotes, pharmacy-data claims, laboratory findings and policy arguments. The patient-data claims still await appraisal of their original records and methods.1716
Does KPV help with long COVID?
The cited studies do not establish a clinical benefit for long COVID. Readers ask about KPV because of its laboratory anti-inflammatory findings. A defined product and patient study would be needed to measure treatment outcomes.1729,1717
Is oral KPV better than an injection?
No cited human study makes that comparison. The animal literature uses several routes, chemical forms and delivery systems, so route comparisons need to keep those differences visible.1717,1730
Does KPV help ulcerative colitis?
Several mouse studies report reduced intestinal inflammation, including work on PepT1 uptake. The formulations range from peptide in drinking water to engineered carriers. No exact-KPV patient trial establishing benefit for ulcerative colitis was found among the sources behind this chapter.1720,1738,1739,1717
Has it been tested in athletes?
The accessible Brazilian sports review names KPV within a broad discussion of peptide use. Its abstract reports no KPV trial or exposure cohort in athletes.1755
Model findings
How might KPV affect inflammation?
Cell and animal experiments report changes in NF-kB signalling and inflammatory mediators. PepT1 is studied as a route of peptide uptake. The signalling target and response depend on the tested cells and peptide form.1720,1721,1722
Does it work like alpha-MSH?
KPV shares alpha-MSH’s last three amino acids. Several experiments observed fragment activity without the usual melanocortin-receptor binding or signalling; other cell systems reported different receptor-related responses. The studies leave the mechanism unresolved across models.1723,1724,1725,1722
Can KPV get through skin?
Researchers measured KPV transport across removed human skin after microporation, electrical delivery or both. Those methods change the delivery conditions. Absorption through living skin needs a defined formulation and a human study.1719,1712
Does a study of a special dressing apply to a plain cream?
A dressing, gel or carrier can change where the peptide goes and how long it stays there. Some carriers also have biological activity. Findings need to stay tied to the tested formulation, comparison groups and model.1712,1716
Can KPV prevent colon cancer?
One mouse study found lower tumor burden in an inflammation-driven colon-cancer model, but no significant protection in PepT1-knockout mice or reduction in tumors in APCMin/+ mice. Its human component measured PepT1 expression in archived colon tissue. The treatment experiments were in mice.1738
Does KPV help with weight loss?
A 2026 Korean paper reports that oral KPV reduced obesity-related measures in mice fed a high-fat diet and suppressed fat-cell development in 3T3-L1 cells. Its abstract leaves the animal dose, duration, group size and exact chemical form unstated.1740
What is the fatty-liver evidence?
A Korean study reports less lipid accumulation in HepG2 cells exposed to KPV at 100 micrograms/mL.1741
What did the skin-pollution study find?
A Korean paper tested 50 micrograms/mL KPV in HaCaT keratinocytes and a 3D skin model exposed to particulate matter. It reports changes in oxidative stress, inflammatory signalling and cell death.1742
Does KPV kill bacteria?
The laboratory findings conflict. An Indian study reported killing of Staphylococcus aureus strains using a Lys-Pro-Val fragment with unstated terminal chemistry; a chemistry study found no antimicrobial activity for Ac-KPV-NH2 and glycoalkylated analogues under its conditions. The experiments used different peptide forms, media and assay methods.1743,1744
What is the yeast-infection evidence?
The Candida-vaginitis experiment used the dimer (CKPV)2 as an intravaginal gel in rats at 0.5, 1 or 2 mg/kg daily, with the strongest reported effect at 2 mg/kg. Methods report 60 female rats overall; Table 2 lists six rats per group.1734
Has KPV been tested for vitiligo?
Chinese studies used KPV-modified liposomes to deliver other active materials in mouse vitiligo models. One carried Nlrp3 shRNA; its human tissue observations investigated disease biology. KPV alone was not administered to patients in that study.1745,1736
What is proKPV?
proKPV is a reactive conjugate designed to release KPV. In DSS-colitis mice, oral proKPV at 0.5 or 2.5 mg/kg daily for seven days improved disease measures, while free KPV at 1 mg/kg showed no benefit in that experiment. Here, free means peptide outside the conjugate; its terminal chemistry was not specified.1730
What did the targeted nanoparticle study add?
Mice with DSS colitis received KPV-loaded nanoparticles with a hyaluronic-acid coating inside a chitosan/alginate hydrogel. The oral study delivered 16 micrograms/kg KPV per day. Methods says five treatment days, while the Figure 7 caption says six; the main figure groups had five mice. The targeted system performed better than the system without the coating.1739
Why was KPV combined with tacrolimus?
Researchers co-assembled KPV with tacrolimus, also called FK506, to study delivery and combined effects in acute and chronic DSS colitis in mice. The nanoparticles generally outperformed the separate treatment groups on inflammation and barrier measures. This was a combination formulation given intravenously.1737
What about mouth sores and wound healing?
The literature includes a KPV-containing mucoadhesive hydrogel for chemotherapy-associated oral mucositis and a diabetic-wound dressing containing KPV and epidermal growth factor. Their abstracts describe preclinical formulation results. Each carrier and additional active ingredient belongs in the interpretation.1746,1747
What is the vascular-calcification research?
A 2024 paper reports preclinical work on a nanodrug assembled from KPV and rapamycin. That abstract does not state the exact KPV form or numeric administration details. It supplies a combination-treatment research lead, with patient outcomes still untested.1748
Has KPV been studied after brain injury?
In mice with experimentally induced brain injury, one intraperitoneal dose of alpha-MSH(11-13) acetate at 1 mg/kg, given 30 minutes after injury, reduced lesion size and apoptotic neurons at 24 hours. The neurological score was not significantly different. The exact terminal chemistry was not independently stated.1749
What did the eye-wound study show?
A rabbit corneal-wound study tested topical alpha-MSH(11-13) and reported faster healing. At 60 hours, the abstract reports complete re-epithelialization in eight of eight corneas treated with KPV or the sodium-nitroprusside comparator. It does not fully explain allocation across the tested concentrations or identify the fragment's terminal chemistry.1750
Can KPV change appetite?
In food-deprived rats given interleukin-1 into the brain, an alpha-MSH(11-13) fragment prevented the loss-of-appetite response at 5.0 pmol but not at 0.5 pmol. The experiment used an intracerebroventricular route.1751
What is the insulin-secretion finding?
An older Japanese experiment reported that alpha-MSH(11-13) inhibited insulin secretion at 10^-9 M in HIT-T15 tumor cells. The abstract leaves exposure duration, replicate count and terminal chemistry unstated.1752
Why does one gut study mention teduglutide?
That mouse study tested cysteine-modified KPV with a hyaluronate and teduglutide nanoparticle platform. It combined mucosal-healing and immunomodulatory approaches. The effect belongs to the full system, including the modified peptide and additional drug.1758
Safety and product quality
Does having only three amino acids make it safe?
Peptide length is one part of the assessment. The July discussion also identified impurities, aggregation, route and final formulation as relevant factors. The human risk has not been measured for a defined KPV product.1712,1716
What does a 99% purity result tell me?
It describes the measured proportion in that particular assay. Peptide amount, water, counterion content and sterility require separate measurements. FDA’s nomination review shows why those results must be distinguished.1712
Were online products independently checked?
A manufacturer told the July committee that seven research-market KPV products had major content and purity problems. The underlying test report, selection method and assay validation are unverified in the cited material.1716
Do zero reported side effects settle the safety question?
The FDA briefing’s search ended in December 2025 and found no KPV reports. Spontaneous reporting does not capture every exposure or event. Human studies with defined products and follow-up are needed to measure risk.1712
Is there a tested human dose or half-life?
FDA’s review found no human pharmacokinetic or pharmacodynamic study for the evaluated forms. The nomination proposed a 0.1% cream or gel, but did not define the final formulation. Those details are not enough to define a human dosing schedule.1712,1716
How long can a KPV product be stored?
Shelf-life depends on the chemical form, formulation, packaging and validated stability data. The storage periods in FDA’s review came from suppliers and certificates for particular materials, so they apply only to those materials.1712
Can KPV affect mood?
Public post titles ask about anhedonia, or loss of enjoyment. No cited controlled KPV study measured mood outcomes. How often such changes occur, and whether the peptide causes them, are unknown.1729,1717
Has KPV been studied during pregnancy or breastfeeding?
FDA’s review did not identify reproductive or developmental toxicity studies, and no cited study administered the peptide during pregnancy or breastfeeding. These are evidence gaps.1712,1717
Have analytical studies checked retail KPV products?
One HPLC study validated measurements using KPV standards and skin homogenates and identified a breakdown product under stress. It was an analytical-method study. The regional corpus contained no chemical testing of consumer KPV products.1753,1754
What do the animal safety experiments show?
The KPV-FK506 paper reports cell-viability, hemolysis and organ-histology assessments for its combination formulation. It states that the longest animal use was two months and calls for longer repeated-use testing.1737
Are interactions with medicines known?
No cited administered-human exact-KPV study measured interactions with medicines. Papers combining KPV with tacrolimus, rapamycin or other drugs test specific preclinical formulations. They do not provide interaction rates for ordinary KPV use in patients.1717,1737,1748
What about toxicity reported for lipidated KPV fragments?
The Indian papers tested a palmitoylated alpha-MSH(11-13) analogue and a gold-nanoparticle conjugate in laboratory systems. The 2022 article has a linked correction whose substantive text was not read for this chapter. Those results stay identified by derivative and experiment.1759,1760,1761
FDA status
What did the FDA committee vote?
On July 23, 2026, the committee voted 8 yes, 6 no and 1 abstention for KPV free base, then gave the same vote for KPV acetate. These were recommendations to FDA about the 503A bulks list.1716
Did that vote approve KPV?
No. Advisory recommendations are considered during FDA’s rulemaking process. Drug approval, a final 503A listing and conditional interim enforcement policy are separate decisions.1716,1726
Is KPV currently in Category 2?
FDA’s current safety page lists it under substances nominated but withdrawn. The currently linked May 14, 2026 category document does not name KPV. Older descriptions of Category 2 need to be dated.1727,1728
Why did FDA staff oppose adding it?
The staff review identified uncertain chemical naming, missing finished-formulation data and insufficient human safety and effectiveness evidence. The committee later voted in favour of recommending both forms. FDA’s final decision follows its rulemaking process.1712,1716,1726
What can the patents tell us?
The Chinese synthesis patent describes COCH3-Lys-Pro-Val-NH2. Russian, Japanese and Korean publications belong to one antimicrobial-lipopeptide patent family. These records document chemical descriptions and applicant claims; clinical outcomes require study data.1756,1757
